N-Acetyl Epitalon Amidate is the terminus-protected form of Epitalon (Ala-Glu-Asp-Gly), incorporating N-terminal acetylation and C-terminal amidation to maximise metabolic stability. This dual terminus modification follows the same stabilisation strategy applied to NA Semax Amidate and NA Selank Amidate — blocking aminopeptidase attack at the N-terminus and carboxypeptidase attack at the C-terminus to extend the compound's effective half-life in biological media.

Epitalon (Ala-Glu-Asp-Gly) is the synthetic tetrapeptide bioregulator developed by Khavinson at the Saint Petersburg Institute of Bioregulation and Gerontology as the minimal active sequence from Epithalamin — a bovine pineal gland extract. The standard Epitalon sequence has a free N-terminal amine at Ala1 that is susceptible to aminopeptidase cleavage, and a free C-terminal carboxylate at Gly4 that is susceptible to carboxypeptidase processing. N-Acetyl Epitalon Amidate addresses both vulnerabilities simultaneously.

The stability advantage of N-Acetyl Epitalon Amidate is research-significant for assays lasting more than 6 hours — telomerase activity assays (TRAP protocol, 2-24 hour incubation), gene expression studies (RT-PCR for AANAT, CLOCK, BMAL1 — 12-24 hours), and cell proliferation studies (48-72 hours). In serum-containing biological media at 37°C, the N-acetyl and C-amide modifications maintain more consistent effective concentrations throughout extended incubation periods compared to standard Epitalon.

The central pharmacophore — Ala-Glu-Asp-Gly — is preserved identically. Neither N-acetylation nor C-amidation alters the central sequence proposed to interact with chromatin-associated proteins or transcription factors in Khavinson's epigenetic mechanism hypothesis. N-Acetyl Epitalon Amidate comparative stability research: to directly measure the stability advantage of the doubly-protected form, prepare N-Acetyl Epitalon Amidate and standard Epitalon at 1µM in three biological matrices: (1) human serum at 37°C, (2) complete cell culture medium (DMEM + 10% FBS) at 37°C, and (3) PBS pH 7.4 at 37°C as an abiological control. At 0, 2, 4, 8, 12, and 24 hours, remove aliquots and measure intact peptide concentration by LC-MS/MS using multiple reaction monitoring (MRM) transitions specific to the intact sequence. Plot remaining intact peptide versus time for each condition. The ratio of half-lives (N-Acetyl Epitalon Amidate half-life / Epitalon half-life) directly quantifies the protection factor conferred by dual terminus modification in each biological matrix.

For telomerase research using the more stable form: seed human fibroblasts (IMR-90 or WI-38) at passage 30-35, approaching replicative senescence, in 6-well plates. Treat with N-Acetyl Epitalon Amidate (1nM-100nM) or vehicle for 72 hours. Harvest cells and measure telomerase activity by the TRAP (Telomeric Repeat Amplification Protocol) assay — heat-inactivated cell extract serves as the negative control; TSR8 synthetic template serves as the positive control for PCR efficiency. Telomerase activity is expressed as TRAP product intensity relative to the internal standard.
